Overview
Peginterferon alfa-2a + vitamin D is a combination therapy investigated by Ziv Hospital for the treatment of chronic hepatitis B (CHB). The regimen consists of peginterferon alfa-2a (a pegylated form of recombinant interferon alfa-2a) administered via subcutaneous injection and vitamin D (typically cholecalciferol) administered orally. Peginterferon alfa-2a acts as an antiviral and immunomodulatory agent by binding to the type I interferon receptor (IFNAR), triggering a cascade of intracellular signaling that inhibits viral replication and enhances the host immune response. Vitamin D is included as an adjuvant immune-modulator; it acts as an agonist at the vitamin D receptor (VDR), which is expressed on various immune cells, potentially enhancing the clearance of the hepatitis B virus and improving sustained virologic response (SVR) rates. This combination was specifically studied in clinical trials to determine if vitamin D supplementation could overcome the relatively low response rates seen with interferon monotherapy in certain HBV genotypes.
