Overview

**Pegvorhyaluronidase alfa + gemcitabine** is an investigational combination therapy consisting of pegvorhyaluronidase alfa (PEGPH20), a pegylated recombinant human hyaluronidase, and gemcitabine, a nucleoside analog and antimetabolite chemotherapeutic. PEGPH20 enzymatically depletes hyaluronan, a major component of the tumor stroma, thereby enhancing local drug delivery and facilitating the uptake of cytotoxic agents such as gemcitabine. This mechanism was developed to address the dense extracellular matrix present in certain high-hyaluronan (HA) tumors, notably metastatic pancreatic ductal adenocarcinoma (PDAC). The combination aimed to increase efficacy by improving the penetration and absorption of chemotherapy within tumor tissue.
