Overview

Pegylated endostatin (PEG-ENDO) is a mono-PEGylated recombinant human form of endostatin, an endogenous protein fragment derived from type XVIII collagen. Endostatin acts as a potent inhibitor of angiogenesis by suppressing vascular endothelial growth factor A (VEGF-A) expression and interfering with multiple signaling pathways involved in endothelial cell migration and proliferation, including inhibition of matrix metalloproteinase 2 (MMP2), matrix metalloproteinase 9 (MMP9), α5β1-integrin-mediated signaling, and Wnt/β-catenin pathways[3][5]. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by preventing microtubule depolymerization. Carboplatin is an organoplatinum alkylating agent that induces DNA cross-linking, leading to apoptosis in rapidly dividing cells[2]. The combination of these three agents has been investigated for the treatment of advanced non-small cell lung cancer (NSCLC), leveraging antiangiogenic effects alongside cytotoxic chemotherapy[5].
