pegylated interferon + tenofovir alafenamide is a combination therapeutic regimen investigated for the treatment of chronic hepatitis B (CHB). It consists of a pegylated form of interferon-alpha, which acts as an immunomodulator and antiviral agent by binding to the Type I interferon receptor (IFNAR), and tenofovir alafenamide (TAF), a next-generation nucleotide reverse transcriptase inhibitor (NRTI). TAF is a prodrug that delivers the active metabolite tenofovir diphosphate into hepatocytes more efficiently than tenofovir disoproxil fumarate, thereby inhibiting HBV DNA polymerase with lower systemic exposure and reduced bone and renal toxicity. The combination is primarily developed and studied by institutions such as the Shanghai Public Clinical Center to achieve a functional cure, defined by the loss of hepatitis B surface antigen (HBsAg), by combining potent viral suppression with immune system stimulation.
