Pegylated interferon alfa-2a + entecavir is a combination therapy used for the treatment of chronic hepatitis B (CHB). Pegylated interferon alfa-2a is a long-acting interferon that acts as an immunomodulator and antiviral agent by binding to the type I interferon receptor (IFNAR), triggering the JAK-STAT signaling pathway and inducing the expression of antiviral genes. Entecavir is a guanosine nucleoside analogue that potently inhibits the hepatitis B virus (HBV) polymerase (reverse transcriptase) by competing with the natural substrate deoxyguanosine triphosphate, thereby blocking viral DNA synthesis. This specific combination and sequential regimen have been studied by Huashan Hospital in multicenter prospective trials to evaluate their ability to induce HBsAg seroclearance in patients who have already achieved viral suppression under long-term entecavir monotherapy.
