Overview

This is a **combination regimen** of four agents used for the treatment of **chronic hepatitis C virus (HCV) infection**: - **Pegylated interferon lambda** is a type III interferon engineered with PEGylation to prolong its half-life and reduce extrahepatic toxicity. It stimulates the JAK/STAT pathway, promoting antiviral, antiproliferative, and immunomodulatory effects. - **Daclatasvir** is a direct-acting antiviral (DAA) functioning as an **NS5A inhibitor**, interfering with HCV replication and assembly. - **Asunaprevir** is an **NS3/4A protease inhibitor**, blocking viral polyprotein processing essential for replication. - **Ribavirin** is a nucleoside analogue with broad-spectrum antiviral activity, often used in combination with interferons and/or DAAs for HCV. This quadruple combination targets multiple steps in the HCV life cycle and is investigated for its efficacy in difficult-to-treat patient populations, especially prior null or partial responders and patients with genotype 1 and 4 infections. It is not a fixed-dose product; rather, the combination is used in clinical protocols. Data show high rates of sustained virologic response (SVR12) in HCV patients who had previously failed pegylated interferon/ribavirin therapy[1][3]. This approach aims to maximize antiviral potency and minimize resistance development.
