Pegylated liposomal doxorubicin (PLD) + ifosfamide is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic soft tissue sarcomas. PLD is a formulation of doxorubicin encapsulated in liposomes with attached polyethylene glycol (PEG) molecules, which increases circulation time, reduces drug leakage, and enhances tumor targeting while reducing cardiotoxicity. Ifosfamide is an alkylating agent that interferes with DNA replication and transcription of RNA, leading to cell death. The combination leverages the antitumor effects of both agents while potentially offering a more favorable toxicity profile compared to conventional doxorubicin combinations.
