Overview

Pegylated liposomal doxorubicin + vinorelbine is a combination chemotherapy regimen used primarily in the treatment of metastatic breast cancer. Pegylated liposomal doxorubicin is a formulation of the anthracycline antibiotic doxorubicin encapsulated in polyethylene glycol-coated (pegylated) liposomes, which enhances drug delivery to tumors and reduces cardiotoxicity. Doxorubicin acts mainly by intercalating DNA and inhibiting topoisomerase II, leading to DNA damage and apoptosis in rapidly dividing cells. Vinorelbine is a semi-synthetic vinca alkaloid that inhibits microtubule assembly, disrupting mitosis and inducing cell death. The combination has demonstrated activity as a salvage regimen for patients with metastatic breast cancer who have been previously treated with other chemotherapies, showing manageable toxicity profiles dominated by neutropenia and mucositis[1][2][3][4].
