Drug intelligence / Profile preview
pelareorep + oxaliplatin + irinotecan + leucovorin + fluorouracil is an investigational combination regimen consisting of the oncolytic virus pelareorep (Reolysin) and the standard-of-care chemotherapy regimen FOLFIRINOX (or modified FOLFIRINOX). Pelareorep is a naturally occurring, non-genetically modified double-stranded RNA reovirus that selectively infects and replicates within cancer cells with activated RAS signaling pathways, leading to tumor cell lysis and the release of tumor-associated antigens. This process stimulates a systemic, proinflammatory immune response, priming the tumor microenvironment for immunologic destruction by recruiting and expanding tumor-infiltrating lymphocytes (TILs). The chemotherapy backbone, FOLFIRINOX, consists of oxaliplatin (a platinum-based DNA alkylating agent), irinotecan (a topoisomerase I inhibitor), leucovorin (folinic acid, a biochemical modulator of fluorouracil), and fluorouracil (5-FU, an antimetabolite). The combination is being developed by Oncolytics Biotech, primarily for the treatment of newly diagnosed metastatic pancreatic ductal adenocarcinoma (PDAC) as part of the GOBLET clinical trial.
