Pembrolizumab + abiraterone + prednisone is a combination therapy evaluated primarily for the treatment of metastatic castration-resistant prostate cancer (mCRPC) in patients who are chemotherapy-naïve. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting programmed death 1 (PD-1), thereby enhancing anti-tumor immune response. Abiraterone is a small molecule inhibitor of CYP17A1, leading to decreased androgen biosynthesis, while prednisone is a synthetic glucocorticoid used to mitigate side effects of abiraterone and provide additional anti-cancer and anti-inflammatory effects. This combination has demonstrated antitumor activity and acceptable safety in clinical studies.
