Overview

Pembrolizumab + ADG106 is a combination of two immunotherapy agents under clinical investigation for the treatment of advanced or metastatic solid tumors and hematological malignancies, including triple-negative breast cancer (TNBC). Pembrolizumab is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2 to enhance anti-tumor immune responses. ADG106 is a fully human, ligand-blocking, agonistic anti-CD137 IgG4 monoclonal antibody developed using Adagene’s NEObody platform. CD137 (also known as 4-1BB) is a member of the tumor necrosis factor receptor family that stimulates T-cell proliferation and survival when activated. The combination aims to synergistically activate immune cells by both releasing inhibitory checkpoints (via pembrolizumab) and providing co-stimulatory signals (via CD137 activation by ADG106), potentially resulting in enhanced anti-tumor activity[1][2][3][4][5][6][7].
