Pembrolizumab + AE37 peptide vaccine is an investigational combination therapy for metastatic triple-negative breast cancer (TNBC). Pembrolizumab is a monoclonal antibody immune checkpoint inhibitor that blocks PD-1, leading to the activation of T-cell-mediated immune responses against tumors. AE37 is a therapeutic peptide vaccine derived from the HER2 protein combined with the Ii-Key motif, designed to activate CD4+ T-helper cells and enhance immune recognition of tumor cells expressing HER2. The rationale for the combination is that AE37 may prime the immune system to increase responsiveness to pembrolizumab by expanding the population of immune-activated tumors. This combination is currently under investigation in phase II clinical trials for TNBC, aiming to leverage complementary mechanisms to improve outcomes in patients not responsive to monotherapy[1][3][5][9].
