Overview

This is a combination regimen consisting of three drugs: - **Pembrolizumab** is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells, blocking its interaction with PD-L1 and PD-L2. This releases the inhibition of immune responses against tumor cells, making it an immune checkpoint inhibitor. - **Albumin-bound paclitaxel** (nab-paclitaxel; commonly known by the brand name Abraxane) is a nanoparticle formulation of paclitaxel bound to albumin. It stabilizes microtubules and inhibits their disassembly during cell division, leading to apoptosis in rapidly dividing cancer cells. - **Cisplatin** is a platinum-based chemotherapeutic agent that forms DNA crosslinks and induces apoptosis in cancer cells. The combination has been studied as induction therapy for various advanced cancers including triple-negative breast cancer (TNBC), head and neck squamous cell carcinoma, and others. The rationale for combining these agents includes synergistic antitumor effects from immunotherapy (pembrolizumab) with cytotoxic chemotherapy (albumin-bound paclitaxel and cisplatin)[1][6].
