Overview

Keytruda-Roger Williams Medical Center-prostate cancer refers to a combination therapy regimen consisting of the PD-1 checkpoint inhibitor pembrolizumab (Keytruda) and autologous anti-PSMA designer T cells. Developed at Roger Williams Medical Center, the designer T cells are a second-generation chimeric antigen receptor (CAR) T-cell therapy engineered to target Prostate-Specific Membrane Antigen (PSMA), a protein highly expressed on the surface of prostate cancer cells. Pembrolizumab is a monoclonal antibody that blocks the Programmed Cell Death Protein 1 (PD-1) receptor, thereby preventing the inhibition of T-cell activity. In this combination, pembrolizumab is utilized to potentially enhance the efficacy, persistence, and infiltration of the CAR-T cells by mitigating the immunosuppressive effects of the tumor microenvironment. This regimen has been investigated in Phase Ia/Ib clinical trials for patients with advanced, hormone-refractory (castration-resistant) prostate cancer.
