Pembrolizumab + axitinib is a combination therapy that targets two different pathways in cancer treatment. Pembrolizumab is a humanized monoclonal antibody that blocks PD-1 and PD-L1 interaction, enhancing and prolonging immune response to the tumor microenvironment. Axitinib is a selective tyrosine kinase inhibitor of VEGF receptors that targets tumor angiogenesis. This combination was approved by the FDA in April 2019 for the first-line treatment of patients with advanced renal cell carcinoma (RCC). In clinical trials, the combination significantly improved overall survival, progression-free survival, and objective response rate compared to sunitinib, with benefits observed across all risk groups regardless of PD-L1 expression.
