**Pembrolizumab + chemoradiotherapy** is a combination regimen in which pembrolizumab, a humanized monoclonal antibody targeting PD-1, is administered alongside standard chemoradiotherapy (CRT). Pembrolizumab blocks the PD-1/PD-L1 interaction to enhance T-cell–mediated antitumor activity. The combination has demonstrated significant survival benefit as both initial and concurrent treatment in high-risk locally advanced cervical cancer and locally advanced, unresectable stage III non–small cell lung cancer (NSCLC). High-level evidence includes phase III and phase II trials showing a statistically significant and clinically meaningful improvement in overall survival and progression-free survival versus CRT alone, notably in cervical cancer and stage III NSCLC[1][2].
