Overview
This combination therapy consists of the immune checkpoint inhibitor pembrolizumab (Keytruda) administered alongside a chemotherapy backbone of cisplatin (or carboplatin) and docetaxel. It is currently being evaluated by Istituti Clinici Scientifici Maugeri SpA in the Phase 2 NeoPeSino trial (NCT05925491) as a neoadjuvant and adjuvant treatment for locally advanced sinonasal undifferentiated carcinoma (SNUC). Pembrolizumab is a humanized monoclonal antibody that blocks the interaction between the PD-1 receptor on T cells and its ligands (PD-L1 and PD-L2), thereby enhancing the anti-tumor immune response. Cisplatin and docetaxel provide cytotoxic effects by inducing DNA damage and inhibiting microtubule depolymerization, respectively, aiming to reduce tumor burden and improve outcomes in this aggressive and rare malignancy.
