Pembrolizumab + denosumab is a combination of two monoclonal antibody drugs used primarily in oncology. Pembrolizumab is an immune checkpoint inhibitor that targets programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2 to enhance T-cell mediated anti-tumor immunity. Denosumab is a fully human IgG2 monoclonal antibody that targets receptor activator of nuclear factor kappa-B ligand (RANKL), inhibiting osteoclast formation and function to reduce bone resorption. This combination has been investigated for use in patients with advanced solid tumors—especially those with bone metastases or metastatic melanoma—to both enhance anti-tumor immune responses and prevent skeletal-related events[2][3][4][6][8].
