Overview

A combination therapy consisting of pembrolizumab, a PD-1 checkpoint inhibitor, and eftilagimod alfa, a soluble LAG-3 protein and MHC Class II agonist. This combination is being evaluated in the EFTISARC-NEO Phase II trial, led by the Maria Sklodowska-Curie National Research Institute of Oncology, as a neoadjuvant treatment for resectable, non-metastatic soft tissue sarcoma in conjunction with radiotherapy. Eftilagimod alfa acts as an antigen-presenting cell (APC) activator, binding to MHC Class II molecules to trigger the activation of CD8+ and CD4+ T cells, dendritic cells, NK cells, and monocytes, thereby enhancing the anti-tumor immune response initiated by pembrolizumab's blockade of the PD-1/PD-L1 pathway.
