Pembrolizumab + interferon alfa-2b is a combination of two immunotherapeutic agents used primarily in the treatment of advanced melanoma and other cancers. Pembrolizumab is a humanized monoclonal antibody that targets programmed cell death protein 1 (PD‑1) on T cells, blocking its interaction with PD-L1 and PD-L2, thereby enhancing the immune system’s ability to detect and destroy cancer cells. Interferon alfa‑2b is an immunomodulatory cytokine that activates immune effector cells such as CD8+ T cells, dendritic cells, and natural killer (NK) cells, while also upregulating PD-L1 expression on tumor and immune cells. The combination aims to synergistically enhance anti-tumor immunity by both priming the immune response (interferon alfa‑2b) and releasing inhibitory checkpoints (pembrolizumab). This regimen has been investigated in clinical trials for metastatic melanoma, showing promising efficacy with manageable toxicity profiles[2][5][8].
