Overview

Pembrolizumab + mFOLFOX7 is a combination regimen used in oncology clinical trials, particularly for metastatic microsatellite stable (MSS) or mismatch repair-proficient (pMMR) colorectal cancer. Pembrolizumab is a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), thereby enhancing T-cell-mediated immune responses against cancer cells. mFOLFOX7 is a modified FOLFOX chemotherapy regimen composed of oxaliplatin, folinic acid (leucovorin), and fluorouracil; it acts by inhibiting DNA replication and promoting cell death in rapidly dividing tumor cells. This combination is employed to potentiate anti-tumor immune responses that may surpass monotherapy efficacy. Primary investigation and development of this combination has occurred in the context of the KEYNOTE-651 clinical trial and similar phase 1b/2 studies in colorectal cancer.
