This combination therapy consists of pembrolizumab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and MRG003 (becotatug vedotin), an antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR). Pembrolizumab, developed by Merck, acts as a checkpoint inhibitor to enhance the immune system's anti-tumor response. MRG003, developed by Lepu Biopharma, consists of an anti-EGFR humanized IgG1 monoclonal antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a valine-citrulline linker. This combination is being investigated as a neoadjuvant and adjuvant treatment in a Phase II trial conducted by Union Hospital, Tongji Medical College, Huazhong University of Science and Technology for patients with Epstein-Barr virus (EBV)-associated locoregionally advanced nasopharyngeal carcinoma.
