Pembrolizumab + platinum-based chemotherapy is a combination drug regimen used as a first-line treatment for adult patients with unresectable advanced or metastatic malignant pleural mesothelioma and other advanced cancers, including non-small cell lung cancer (NSCLC)[2][1][3]. Pembrolizumab is a humanized monoclonal antibody targeting programmed death receptor-1 (PD-1); it blocks the interaction between PD-1 and its ligands PD-L1 and PD-L2, thereby increasing T-cell activation against tumor cells[3]. The platinum-based chemotherapy includes agents such as cisplatin or carboplatin, which act through DNA damage and inhibition of DNA repair, causing cytotoxicity and apoptosis in cancer cells[3]. This combination leverages immune checkpoint inhibition together with direct cytotoxic antineoplastic activity for enhanced clinical efficacy.
