Overview

Combination of three immune-oncology monoclonal antibodies: **pembrolizumab** (an anti–PD-1 antibody), **quavonlimab** (an anti–CTLA-4 antibody), and **MK-4830** (an anti–ILT4 antibody). Pembrolizumab blocks the programmed cell death-1 (PD-1) receptor, reactivating T cells and enhancing immune response against tumors. Quavonlimab targets cytotoxic T-lymphocyte–associated protein 4 (CTLA-4), further promoting T-cell activation. MK-4830 inhibits immunoglobulin-like transcript 4 (ILT4/LILRB2), which is expressed on myeloid cells and contributes to tumor immune evasion. The combined blockade aims to synergistically enhance anti-tumor immunity in refractory or advanced cancers, particularly microsatellite instability-high/mismatch repair-deficient (MSI-H/dMMR) colorectal cancer, small cell lung cancer, and other advanced solid tumors. The combination is under investigation chiefly in phase 1/2/3 clinical trials.
