Overview

pembrolizumab + tazemetostat is a combination therapy being evaluated as a maintenance treatment for aggressive B-cell non-Hodgkin lymphoma (NHL). The regimen combines pembrolizumab, a humanized monoclonal antibody that targets the programmed death-1 (PD-1) receptor to enhance the immune system's ability to detect and destroy tumor cells, with tazemetostat, an oral small molecule inhibitor of the EZH2 histone methyltransferase. This combination is specifically being studied in patients who have achieved a response following autologous stem cell transplantation (ASCT) or CAR T-cell therapy to overcome immune tolerance and prevent relapse. The Phase II trial (NCT06242834) is led by Northwestern University and targets various aggressive B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL) and mantle cell lymphoma.
