Overview
This drug is a four-agent combination therapy consisting of two immune checkpoint inhibitors (**pembrolizumab**, a PD-1 inhibitor, and **vibostolimab**, a TIGIT inhibitor) along with two cytotoxic chemotherapies (**carboplatin**, a platinum-based DNA crosslinker, and **paclitaxel**, a mitotic spindle inhibitor). The immune checkpoint inhibitors work by augmenting anti-tumor immune responses: pembrolizumab blocks PD-1 on T cells, preventing tumor-induced immune suppression; vibostolimab blocks TIGIT, further enhancing T cell activation. Carboplatin induces DNA damage and cell death, while paclitaxel disrupts microtubule dynamics needed for cell division. The regimen is under investigation for the treatment of various advanced cancers, including head and neck squamous cell carcinoma (HNSCC) and non-small cell lung cancer (NSCLC) in first-line and refractory settings. The combination is designed to optimize anti-tumor effects through synergistic immunotherapy and chemotherapy mechanisms[1][2][4][5].
