CBP501 is a synthetic duodecapeptide that enhances the cytotoxicity of cisplatin by increasing its influx into tumor cells through interaction with calmodulin and by abrogating DNA repair at the G2 checkpoint. When combined with standard chemotherapy agents pemetrexed (an antifolate) and cisplatin (a platinum-based chemotherapeutic), this triplet regimen has been investigated for the treatment of malignant pleural mesothelioma and non-small cell lung cancer. Pemetrexed inhibits folate-dependent enzymes involved in nucleotide synthesis, while cisplatin forms DNA crosslinks leading to apoptosis. The addition of CBP501 aims to sensitize tumor cells to cisplatin without affecting normal cells, potentially improving efficacy over standard doublet therapy[1][3][6]. The combination is administered intravenously in 21-day cycles, often with vitamin supplementation to reduce toxicity[2][5].
