Pemetrexed + prolgolimab is a combination therapy used as a first-line treatment for advanced non-squamous non-small cell lung cancer (NSCLC). Pemetrexed is an antineoplastic agent that inhibits folate-dependent enzymes involved in nucleotide synthesis, thereby disrupting DNA and RNA synthesis in rapidly dividing tumor cells. Prolgolimab is an IgG1 anti-PD-1 monoclonal antibody with an Fc-silencing 'LALA' mutation; it blocks the interaction between the PD-1 receptor on T cells and its ligands PD-L1/PD-L2, enhancing immune-mediated tumor cell killing. The combination has demonstrated improved overall survival and progression-free survival compared to chemotherapy alone, with a favorable safety profile and efficacy independent of PD-L1 status[1][6].
