Pemetrexed + vandetanib is a combination therapy consisting of pemetrexed, an antifolate chemotherapy agent, and vandetanib, a multi-targeted tyrosine kinase inhibitor. Vandetanib inhibits vascular endothelial growth factor receptor (VEGFR), epidermal growth factor receptor (EGFR), and RET signaling pathways, while pemetrexed disrupts folate-dependent metabolic processes essential for cell replication. This combination was investigated primarily for second-line treatment of advanced non-small cell lung cancer (NSCLC).
