Pemetrexed + vinorelbine is a chemotherapy combination regimen that has been studied for the treatment of various cancers, particularly non-small cell lung cancer (NSCLC) and metastatic breast cancer. Pemetrexed is a multi-targeted antifolate that inhibits several folate-dependent enzymes involved in DNA synthesis, including thymidylate synthase (TS), dihydrofolate reductase (DHFR), and glycinamide ribonucleotide formyltransferase (GARFT). Vinorelbine is a semisynthetic vinca alkaloid that works by binding to tubulin in mitotic microtubules, inhibiting cell division. When combined, these agents provide complementary mechanisms of action that target different aspects of cancer cell growth and replication. This combination has shown efficacy in non-small cell lung cancer and metastatic breast cancer in clinical trials.
