pemigatinib + durvalumab

Pemigatinib + durvalumab is a combination therapy that pairs a selective FGFR1-3 inhibitor (pemigatinib) with a PD-L1 blocking antibody (durvalumab). Pemigatinib works by inhibiting fibroblast growth factor receptor (FGFR) signaling, blocking receptor autophosphorylation and downstream FGF/FGFR signaling in FGFR-driven cancers, particularly those with FGFR2 alterations. Durvalumab is a human immunoglobulin G1 kappa (IgG1κ) monoclonal antibody that blocks PD-L1 from binding to PD-1 and CD80, thereby activating immune responses mediated by cytotoxic T cells against tumor cells. The combination of targeted drugs and immunotherapies has demonstrated clinical value in various tumors, and this specific combination is being investigated for biliary tract cancers, particularly those with FGFR2 alterations.
