**Pemigatinib + pembrolizumab** is an investigational combination therapy consisting of pemigatinib, a selective small molecule inhibitor of fibroblast growth factor receptor (FGFR) 1–3, and pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed cell death protein 1 (PD-1). Pemigatinib acts by inhibiting the FGFR pathway, which is implicated in tumor cell proliferation and survival, especially in cancers with FGFR alterations. Pembrolizumab blocks PD-1, releasing the “brakes” on the immune system to augment anti-tumor immune response. The combination has demonstrated preliminary antitumor activity and a manageable safety profile in early-phase trials for advanced malignancies, particularly in patients with FGFR genetic alterations or refractory solid tumors[1][2][3]. Both modalities show potentially synergistic mechanisms, targeting tumor proliferation and immune escape simultaneously.
