This combination therapy regimen is being evaluated in a Phase 2 randomized clinical trial at Xijing Hospital for the perioperative treatment of locally advanced resectable esophageal cancer. The regimen integrates immunotherapy, targeted therapy, and chemotherapy, consisting of **penpulimab** (an anti-PD-1 monoclonal antibody), **anlotinib** (a multi-target tyrosine kinase inhibitor), and a chemotherapy backbone of **nab-paclitaxel** (albumin-bound paclitaxel) combined with either **oxaliplatin** or **lobaplatin**. Penpulimab works by blocking the PD-1/PD-L1 pathway to restore T-cell mediated anti-tumor immunity. Anlotinib provides anti-angiogenic and anti-proliferative effects by inhibiting VEGFR, FGFR, PDGFR, and c-Kit. The chemotherapy components, nab-paclitaxel and platinum agents, induce cytotoxic effects through microtubule stabilization and DNA cross-linking, respectively. The study specifically compares the efficacy and safety of oxaliplatin versus lobaplatin within this multi-drug framework for patients with clinical stage cT2-4N0-3M0 esophageal cancer.
