Penpulimab + oxaliplatin + capecitabine is a chemo-immunotherapy combination regimen being evaluated for the treatment of advanced solid tumors, particularly gastric and gastroesophageal junction (GEJ) cancers. The regimen consists of **penpulimab** (AK105), a humanized IgG1 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor to enhance anti-tumor T-cell activity, combined with the **XELOX** (or CAPOX) chemotherapy doublet. XELOX includes **oxaliplatin**, a platinum-based cytotoxic agent that induces DNA cross-linking, and **capecitabine**, an oral fluoropyrimidine prodrug that is converted to 5-fluorouracil (5-FU) to inhibit DNA synthesis. This combination is often studied as a backbone therapy in clinical trials alongside novel agents such as LM-108 (an anti-CCR8 antibody) to improve outcomes in patients with advanced gastrointestinal malignancies.
