Overview

Pep(4) is a synthetic peptide derived from the Herpes Simplex Virus (HSV) glycoprotein D (gD), specifically representing the 1-17 amino acid sequence of the protein. It functions as an immune checkpoint inhibitor by targeting the Herpesvirus Entry Mediator (HVEM), also known as TNFRSF14. Pep(4) is designed to competitively inhibit the binding of B- and T-lymphocyte attenuator (BTLA) to HVEM. By disrupting the BTLA-HVEM inhibitory complex, Pep(4) aims to restore and enhance T-cell mediated immune responses against tumors. Research has primarily focused on its potential application in melanoma, where it has demonstrated the ability to modulate cytokine production and T-cell activation in patient-derived samples.
