Peposertib + avelumab is a combination of two investigational agents studied for advanced solid tumors. Peposertib (M3814) is a potent, selective small molecule inhibitor of DNA-dependent protein kinase (DNA-PK), a critical enzyme in DNA double-strand break repair by nonhomologous end joining (NHEJ). Avelumab is a monoclonal antibody targeting programmed death-ligand 1 (PD-L1), functioning as an immune checkpoint inhibitor. The combination was evaluated in a phase I trial, demonstrating tolerability up to defined dose levels, but with limited antitumor activity in a heavily pretreated population. Primarily developed and manufactured by Merck and Pfizer (avelumab), the combination is studied as an antineoplastic, immunotherapy regimen seeking to synergize DNA damage and adaptive immune response inhibition[1][3].
