PeptiCRAd-1 + pembrolizumab is an investigational **combination immunotherapy** being evaluated primarily for cancer treatment. PeptiCRAd-1 is a novel clinical vaccine platform that consists of a **conditionally replicating oncolytic adenovirus (VALO-D102) coated with tumor-specific peptides** from cancer-testis antigens NY-ESO-1 and MAGE-A3, designed to stimulate potent cytotoxic T cell responses against tumors. This virus also expresses the immune-stimulatory ligands CD40L and OX40L, further activating the immune response. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting PD-1. The combination aims to harness direct oncolytic activity, **tumor-specific T-cell activation, and checkpoint inhibition** to generate broader and more durable anti-tumor responses, especially in tumors expressing NY-ESO-1 and MAGE-A3. Developed and currently being trialed by Valo Therapeutics, this therapy is under phase I evaluation for melanoma, triple negative breast cancer, non-small cell lung cancer, sarcoma, and colorectal cancer (NY-ESO-1 or MAGE-A3 positive)[1][3][5][7].
