This investigational cancer vaccine consists of autologous dendritic cells pulsed with three HLA-A2-restricted tumor-associated peptides—human telomerase reverse transcriptase (hTERT, TERT572Y), carcinoembryonic antigen (CEA; Cap1-6D), and survivin (SRV.A2)—administered in combination with Poly-ICLC (Hiltonol), a synthetic double-stranded RNA and Toll-like receptor 3 (TLR3) agonist. Developed by researchers at the Medical University of South Carolina under the leadership of Dr. Carolyn Britten, the therapy is designed to treat metastatic or locally advanced unresectable pancreatic adenocarcinoma. The dendritic cells act as professional antigen-presenting cells to prime the immune system, while Poly-ICLC serves as a potent adjuvant to enhance the magnitude and quality of the tumor-specific T-cell response.
