This experimental immunotherapy regimen, developed by the National Cancer Institute (NCI), is designed for patients with recurrent pediatric sarcomas, specifically Ewing sarcoma and alveolar rhabdomyosarcoma. The protocol utilizes a tumor-specific peptide-pulsed antigen-presenting cell (APC) vaccine to prime the immune system against disease-specific fusion proteins, such as EWS-FLI1 (associated with Ewing sarcoma) and PAX3-FKHR (associated with alveolar rhabdomyosarcoma). This vaccination is followed by the administration of recombinant human interleukin-2 (IL-2), a cytokine that acts as a T-cell growth factor to expand and activate tumor-reactive T cells. The regimen may also incorporate autologous T-cell transplantation to enhance the anti-tumor immune response by providing a larger pool of effector cells.
