**Perindopril + indapamide + amlodipine** is an oral, single-pill fixed-dose antihypertensive combination containing an angiotensin-converting enzyme inhibitor, a thiazide-like diuretic, and a dihydropyridine calcium-channel blocker. Perindopril is converted to perindoprilat, which inhibits angiotensin-converting enzyme and reduces angiotensin II formation; indapamide promotes renal sodium and chloride excretion; and amlodipine inhibits L-type calcium channels in vascular smooth muscle to cause vasodilation. The combination is used for essential hypertension, particularly as substitution therapy for patients already controlled on the corresponding component doses. Servier developed the reference product Triplixam, also designated S06593; generic fixed-dose products include Krka's Co-Amlessa and Darnitsa's Perindopres Trio.
