Pertuzumab + paclitaxel is a combination therapy used in the treatment of HER2-positive breast cancer, especially in metastatic and early-stage settings. Pertuzumab is a monoclonal antibody that binds to domain II of the human epidermal growth factor receptor 2 (HER2), preventing dimerization with other HER family receptors and thereby inhibiting downstream signaling that promotes cancer cell growth. Paclitaxel is a taxane chemotherapeutic agent that stabilizes microtubules, interfering with cell division and leading to cancer cell death. The combination exploits the complementary mechanisms of HER2 blockade and cytotoxic chemotherapy, providing enhanced antitumor activity. This combination is often administered with trastuzumab in clinical practice and recommended in guidelines as a well-tolerated, effective regimen for HER2-positive metastatic breast cancer and in some early stages[3][4][1].
