Overview

Pertuzumab + trastuzumab + docetaxel is a three-drug combination regimen used primarily for the treatment of HER2-positive breast cancer, including both metastatic and early-stage (neoadjuvant) settings. Pertuzumab and trastuzumab are recombinant humanized monoclonal antibodies that target different epitopes on the human epidermal growth factor receptor 2 (HER2), providing dual blockade of HER2 signaling. Docetaxel is a cytotoxic chemotherapy agent classified as a taxane, which inhibits cell division by stabilizing microtubules. The combination has demonstrated significant improvements in progression-free survival and overall survival compared to regimens without pertuzumab in clinical trials such as CLEOPATRA and NeoSphere[1][3][4][5][6][9]. This regimen is considered standard first-line therapy for patients with HER2-positive metastatic breast cancer who have not received prior anti-HER2 therapy or chemotherapy for metastatic disease.
