Drug intelligence / Profile preview
Poherdy + Hercessi + docetaxel is a combination regimen consisting of two biosimilar monoclonal antibodies and a taxane chemotherapy agent, used for the treatment of HER2-positive breast cancer. Poherdy (pertuzumab-dpzb) is an interchangeable biosimilar to Perjeta (pertuzumab) that targets subdomain II of the human epidermal growth factor receptor 2 (HER2), preventing HER2 heterodimerization with other HER family receptors. Hercessi (trastuzumab-strf) is a biosimilar to Herceptin (trastuzumab) that binds to subdomain IV of HER2, inhibiting ligand-independent HER2 signaling. Docetaxel is a microtubule inhibitor that promotes microtubule assembly and inhibits disassembly, leading to cell cycle arrest and apoptosis. Together, the dual HER2 blockade by pertuzumab-dpzb and trastuzumab-strf, combined with the cytotoxic effects of docetaxel, provides synergistic anti-tumor activity and enhances antibody-dependent cell-mediated cytotoxicity (ADCC) against HER2-overexpressing cancer cells. This regimen is indicated for the neoadjuvant, adjuvant, and first-line metastatic treatment of HER2-positive breast cancer.
