Pexastimogene devacirepvec (Pexa-Vec, JX-594) is an oncolytic virus therapy based on a vaccinia virus backbone that has been genetically engineered to selectively replicate in and destroy tumor cells. It achieves cancer selectivity by deactivating its thymidine kinase gene and expresses granulocyte-macrophage colony-stimulating factor (GM-CSF) to stimulate systemic anti-tumor immunity. Socazolimab (formerly known as ZKAB001) is a recombinant human anti-PD-L1 monoclonal antibody that blocks the interaction between PD-L1 and its receptor PD-1, thereby enhancing T-cell mediated immune responses against tumors. The combination of Pexa-Vec with Socazolimab is being investigated for synergistic effects in treating cancers such as metastatic melanoma and extensive-stage small-cell lung cancer[1][3][5][8].
