PF-03084014 + docetaxel is a combination therapy consisting of PF-03084014 (also known as nirogacestat), a selective, small-molecule γ-secretase inhibitor, and docetaxel, a well-established taxane chemotherapeutic agent. PF-03084014 inhibits the γ-secretase complex, thereby blocking NOTCH receptor activation and downstream signaling pathways implicated in tumor growth and survival. Docetaxel disrupts microtubule function during cell division, leading to apoptosis in rapidly dividing cancer cells. This combination is under investigation for synergistic antitumor effects in advanced solid tumors.
