Overview

**PF-04217329 + latanoprost** is a dual combination ophthalmic therapy comprising *taprenepag isopropyl* (PF-04217329), a selective prostaglandin EP2 receptor agonist, and *latanoprost*, a prostaglandin F2α analog. The drug is primarily investigated for its ability to lower intraocular pressure (IOP) in patients with primary open-angle glaucoma or ocular hypertension. Taprenepag isopropyl promotes aqueous humor outflow via EP2 receptor activation, while latanoprost increases uveoscleral outflow through prostaglandin F receptor agonism. Clinical and model-based meta-analyses indicate a synergistic IOP-lowering effect when combined, exceeding the efficacy of either agent as monotherapy. The combination is administered as separate ophthalmic solutions (unfixed combination) and has not reached the market as a fixed-dose product[1][3].
