Overview

PF-04856884 + axitinib is an investigational combination therapy comprising **PF-04856884 (CVX-060)**, a monoclonal antibody that selectively inhibits **angiopoietin-2 (Ang2)**, and **axitinib**, a small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFRs), primarily **VEGFR-1, VEGFR-2, and VEGFR-3**. The purpose of this combination is to disrupt tumor blood vessel stability and angiogenesis via dual blockade of the angiopoietin/Tie2 and VEGF receptor pathways. Developed for metastatic renal cell carcinoma, it was tested in patients previously treated with one systemic therapy. The program was terminated in phase 2 due to tolerability concerns, including unexpected arterial and venous thrombotic events[1][3][5].
