Overview

PF-06647020 + avelumab is an investigational combination therapy for cancer, currently studied in advanced solid tumors, including platinum-resistant ovarian cancer. PF-06647020 (cofetuzumab pelidotin) is an antibody-drug conjugate (ADC) composed of a humanized anti-protein tyrosine kinase 7 (PTK7) monoclonal antibody linked to the auristatin microtubule inhibitor payload Aur0101 via a cleavable linker. It targets PTK7-expressing tumor cells, delivering the cytotoxic agent directly to induce mitotic arrest and apoptosis. Avelumab is a fully human IgG1 monoclonal antibody that blocks programmed cell death ligand 1 (PD-L1), preventing its interaction with PD‑1 and B7 receptors on T cells and antigen-presenting cells, thereby enhancing antitumor immune responses. The combination aims to leverage both direct cytotoxicity against tumor cells and immune-mediated antitumor activity[2][3][6][8].
