PF-06747143 + azacitidine is a **combination therapy** investigated for the treatment of **acute myeloid leukemia (AML)**. PF-06747143 is a novel humanized IgG1 monoclonal antibody targeting the **chemokine receptor CXCR4** on AML cells, inhibiting CXCL12-induced migration, mobilizing malignant cells from bone marrow to peripheral blood, and mediating cytotoxicity through antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Azacitidine is a **hypomethylating agent** that incorporates into DNA and inhibits DNA methyltransferase, leading to DNA hypomethylation and cytotoxicity in abnormal hematopoietic cells. This combination was assessed in a clinical trial in adult patients with relapsed or refractory AML to determine safety, pharmacokinetics, pharmacodynamics, and dose-limiting toxicities. The combination is not approved for use outside of clinical trials.
