Drug intelligence / Profile preview
PF-06747143 + decitabine is an investigational drug combination for hematologic malignancies, especially acute myeloid leukemia (AML). PF-06747143 is a humanized IgG1 monoclonal antibody that antagonizes the chemokine receptor CXCR4. It inhibits CXCL12-mediated signaling, blocks malignant cell migration/homing to the bone marrow, and induces tumor cell death via Fc-mediated effector function. Decitabine is a small molecule hypomethylating agent, inhibiting DNA methyltransferase, and used to promote re-expression of silenced tumor suppressor genes. Preclinical and early clinical studies show that PF-06747143 can synergize with standard agents like decitabine for enhanced anti-leukemic activity, particularly in chemoresistant AML models. This combination is being explored for relapsed/refractory and newly diagnosed AML, especially in patients unsuitable for intensive induction chemotherapy[2][3][4][6].
